Is there a way to run AC from USB drive? Or putting the data files on USB drive and running AC installed on a computer?

You could copy the databases onto the flash drive, but as to a standalone Amazing Charts portable application, I don't think that will work. Even if it did, I would be reluctant to use it given the speed of a standard USB flash drive. Perhaps if you provided us some more information regarding your situation, we could recommend other alternatives. If I had to run it off a flash drive, I would use a USB 3.0 SuperTalent ST3U32EDK at minimum.

Also USB flash drives have a higher failure rate than other forms of storage media, are fragile & easily misplaced, so may not be optimal for patient records.

Running SQL Server databases from a USB flash drive is completely unsupported by Microsoft afaik. Furthermore, you risk database corruption should someone accidentally pull the drive out of the computer while the database is being written to.

Believe me, this is a bad idea.
